ABSTRACT vii The study is aimed at evaluating the antioxidant activity of the Allium sativum sample, its antiinflammatory and hepatoprotective effect in Albino rats. The antioxidant activity was determined by DPPH RSA, ALP activity and nitric oxide I A. 200mg/kg b.w of Allium sativum showed similar antioxidant activity when compared to lOOmg/kg vit. C. Cone. The Allium sativum concentrations (mg/dl) showed a significant decrease p<0.05 in antioxidant activity and 200mg/ml of Allium sativum showing the highest antioxidant activity in Allium sativum 'concentrations. The anti-inflammatory effects of Allium sativum was evaluated by acute inflammatory model. Reduction in Paw edema size was recorded in minutes. The administration of extract at doses of 200, 400 and 600mg/kg body weight to the rats resulted in a significant 'decrease (p< 0.05) compared to control group. EEAS was able to show highest significant decrease (p<0.05) in egg albumin induced paw edema at concentration of 400mg/kg at the end of |240 minutes, (i.e 0.45±0.04 [41.6%]) when compared with negative control saline. The percentage inhibition exhibited by 200mg/kg, 400mg/kg, and 600mg/kg of EEAS (31.2%, 41.6% and 32.5% respectively) was similar to that of piroxicam, which has shown 48.1% of inhibition. The result of the study suggest that Allium sativum posses anti-inflammatory properties. The hepatoprotective activity of Allium sativum was evaluated using Alcohol (vol 47%) to induce toxicity. The observed effect when blood sample was collected and subjected to biochemical assay (AST, ALT, Total bilirubin and conjugate bilirubin) was done using standard kits. The standard drug group and EEAS group showed a significant decrease (p<0.05) when compared to the control group.. In AST and ALT the drug and treatment group showed a significant decrease (*p<0.05) when compared to the control group. After 14 days of induction of hepatotoxicity EEAS was able to show the highest level of decrease in AST and ALT level at 200mg when compared to other EEAS group and was similar To that of silymarin 15mg/kg b.w. This shows the extract posses hepatoprotective properties.
