ABSTRACT Doxorubicin (DOX) is a potent and widely used anthracycline antibiotic for the treatment of cancers. Scientific research have shown evidence of cardiotoxicity arising from clinical use of DOX in treatment of cancer patients which is the major impeding issue pertaining to the clinical application of DOX in treatment of cancer. The present study focused to examine the cardioprotective effect of Clove seed in DOX induced cardio toxicity in rats. Twenty (20) male Wister rats of 180±20g average weight were obtained and allowed to acclimatize for 7 days before commencing treatment. The ratt were grouped into four (n=5). Group 1 (normal control) received normal saline, Group2 (untreated) received weekly dose 20mg/kg of DOX, Group 3 received weekly dose of 20mg/kg DOX plus daily dose of 200mg/kg of clove extract, Group 4 received weekly dose of 20mg/kg DOX plus daily dose of 400mg/kg of clove extract, Animals were sacrificed after 21 days of treatment and blood sample were collected for parameter analysis. Result shows a significant deccrease (P<0.05) in Troponin CK, LDH, SOD and MDA levels in group 4 when compared with group 2, however Troponin, CK, LDH, SOD and MDA level in group 4 were insignificantly different with group 1. There was a significant increase (P<0.05) in Total protein level in group 4 when compared with group 2. group 4 were insignificantly different with group 1 and group 3 in the level of Total protein. It was therefore ascertained that administration of 400 mg/kg body weight dose of Syzygium aromaticum (clove) seed extract has protective effect against oxidative stress and apoptotic damage in experimental model of Doxorubicin- induced cardiac toxicity in rats. These protective potentials ofthe clove seed were attributed to the abundant phytochemicals present in the clove. Further studies with the use of higher animal and higher doses of the plant’s extracts was recommended to ascertain the applicability in use of seed extract of clove for management and treatment of cardotoxicity. Furthermore extensive studies on Clove’s bioavailability, specificity, and underlying mechanisms of action, including nutrigenomic approach and in-depth analysis of Clove’s phytoconstituents-gene interaction was also recommended to enhance the potential clinical use of natural products in management of cardiac diseases.
