ABSTRACT
Lectins are
carbohydrate binding proteins present in most plants. They are toxic,
inflammatory, and resistant to cooking and digestive enzymes and present in
many of our foods. In this study, lectins were extracted from Okazi leaf
(Gnetum africanum), Bitter leaf (Venonia amygdalina), Fluted pumpkin leaf
(Telferia occidentalis), and Oha leaf (Pterocarpus soyauxii). Partial
purification of the lectin extracts was achieved by precipitation with ammonium
sulphate and the extract was tested for hemagglutinin activity using 4%
suspension of human erythrocytes in lectin buffer. Extracts from the above
selected vegetables agglutinated all the blood groups (A, B, AB and 0)
suggesting the presence of non blood type specific lectin in the vegetables.
Since agglutination readily occurred without the need for proteolytic enzyme or
addition of metal ion, the crude extract could be regarded as complete lectin.
The hemagglutinin activity of the vegetables lectin were found decrease when
exposed to processing methods ( oven drying and moist heating). Also the
extract decreased with increase in incubation temperature. Strong and moderate
agglutination was observed in non incubated sample and samples incubated at 10C
and 30C while agglutinations was devoid when subjected to temperature of 70C
and 100C, suggesting that lectin from these vegetables are heat labile and can
be deactivated by moist or dry heating.
