ABSTRACT
This study showed the effect of methanol extract
on Malay Apple leaf{Syzygium malaccense) on Prostate specific Antigen (PSA) and
antioxidant properties in dihydroestestrone - estradiol valarate induced Benign
Prostate Hyperplasia (BPH) in male wistar rat. Thirty male Albino rats
(90g-150g) were randomly divided in 5 groups. Group 1 (Normal control), Group 2
(Disease control), Group 3 (Drug control), Group 4 (Syzygium malaccense lOOmg/Kg),
Group 5 (Syzygium malaccense 200mg/Kg). Benign Prostate Hyperplasia (BPH) was
induced by subcutaneous Injection of 9mg/Kg and 0.9mg/Kg body weight
Dihydrotestosterone and Estradiol valarate both dissolved in olive oil, every
day for 28 days. Group 1 received subcutaneous injection ofthe hormone dose
vehicle (Olive oil) in place ofthe hormone and a dose ofthe same vehicle (olive
oil). Group 3, 4, 5 received subcutaneous injection of the vehicle (olive oil)
and concomitant oral administration of Finasteride (5mg / 70kg), Syzygium
malaccense (lOOmg/Kg), Syzygium malaccense (200mg/Kg) respectively every day
for 28 days. Group 2 received BPH induction and concomitant oral administration
of dose vehicle (which served as placebo) every day for 28 days. There was no
significant increase (P<0.05) in the relative organ weight of the entire
groups when compared to Group 1 (Normal Control). Relative prostate weight and
Prostate Specific Antigen significantly increased (P<0.05) in Group 2
(Disease control) compared to Group 1 (Normal control), Group 3(Drug control),
Group 4 (Syzygium malaccense lOOmg/Kg) and Group 5 (Syzygium malaccense
200mg/Kg) (Group 5). Antioxidant parameters - Superoxide Dismutase (SOD),
Glutathione peroxidase (GPx) increased significantly (P<0.05) except for
Malondialdehyde (MDA). These findings suggest that Syzygium malaccense has
positive effect in management of Benign prostate hyperplasia (BPH) as it
contains bioactive agents and Antioxidants that may be of pharmacological
importance in the management of Benign prostate hyperplasia (BPH).
