ABSTRACT

 The project studied the isolation of thermophilic microorganisms (fungi and bacteria) with Amylase, protease and lipase activity from waste dumpsites. Thirty dumpsite soil samples were screened for fungi and bacteria flora and isolates were screened for enzymes potentials (arnylase, protease and lipase) and the optimum conditions (temperature and pH) for enzyme production. Obtained results show that many fungi and bacteria isolates made up the microbial flora of the dumpsite soils and with varying prevalence of occurrence including Aspergillus niger (80%), Mucor (53.3%), Yeast (86.7%), Aspergillusfun2igatus (43.3%), Penicillum (40.0%), Fusariurn (33.3%) and Aspergillus flavus (. 36.7%). The occurrence and prevalence of the bacteria isolates showed Bacillus subtilis (70.0%), Bacillus lichenforniis (43.3%), Bacillus cereus (56.7%), Pseudomonas aeruginosa (56.7%), Streptomyces species (36.7%), Bacillus brevis (43.3%). The enzyme activity test showed that the fungi which produced all three enzymes were Aspergillus niger, Aspergillus fumigatus and Penicillin while Mucor produced only arnylase and protease, Fusarium produced lipase and cellulose only, while Aspergiliusfiavus produced arnylase, protease, and cellulose. Also the bacteria were able to produce three enzymes. Optimization of the conditions for the enzyme production by thermophiles show that the fungi isolates had optimum temperatures between 60°C and 65°C while the bacteria isolates had between 6.0 and 7.0 with varying rates of enzyme productions. The potentials of the microbial isolates as good sources of enzymes even for commercial uses was recognized.
