ABSTRACT

This study was carried out to determine the incidence of bioflim formation and Methicillin resistance among clinical isolates of Staphylococcus aureus obtained from Mercy Specialist Hospital, Ahiaeke, Umuahia, Abia state. A total of 30 randomly selected patients were included in the study. Clinical samples collected were subjected' to standard bacteriological culture. S. aureus isolates were identified by mannitol fermentation, coagulase positivity and catalase positivity. Antimicrobial susceptibility test was performed on Mueller-hinton agar (MHA) by modified Kirby-Bauer disc diffusion method. Biofilm producers were also detected using the conga red agar method. Out of total 30 clinical samples, S. aureus was isolated in 63.3% (n=19). Among the isolates, 63.1% (n=12) were from male patients whereas 36.8% (n=7) were from female. There was no sex difference in colonization of S. aureus. Out of 19 isolates, 68.4% (n=13) were MRSA. MRSA isolates indicated relatively high rate of resistance to antibiotics Ceftriaxone (84.6%) followed by Erythromycin and Ciprofloxacin with (76.9%) each, then Cloxacillin (69.2%) and Ampicillin (61.5%) respectively. The isolates showed the lowest resistance to Gentamicin (30.8%). Methicillin resistance was determined using the Cefoxitin and Oxacillin disc diffusion methods. Biofilm formation in Staphylococcus aureus isolates using the Congo red agar (CRA) method could detect 10 (52.6%) of Staphylococcus aureus as strong bioflim producers which include 9 (47.4%) MRSA, and 1 (5.3%) MSSA. This study showed a slightly high incidence of biofilm formation and Methicillin resistance among clinical isolates of Staphylococcus aureus obtained from Mercy Specialist Hospital, Ahiaeke, Umuahia, Abia state.
