ABSTRACT
The study was designed to evaluate the hepatoprotective and renal protective activity of Pentaclethra inacrophylla seed. Twenty four wistar rats weighting 100 — 240g were divided into seven groups of four animals each. Group 1 were used as negative control (-ye control), groups 2 served as positive control (+ve control), while group 3-7 were maintained as test group administered different doses of Pentacleihra niacrophylla seed extract. Hepatorenaltoxicity was induced with paracetamol in all groups, after fourteen days, the animals were sacrificed and their blood collected through cardiac puncture, the blood was separated and their serum were collected for biochemical estimation. The result obtained were presented as rnean±S.D. Antioxidant studies carried out to determine the DPPH radical scavenging activity showed a significant P 0.05 decrease ranging from 14.76±0.55 to 86.50±5.24 at a maximum concentration of 200mg/mI when compared with the standard drug activity with value of 97.60±0.85 at 100mg/mI concentration .Anti-lipid peroxidation activities showed a significant P 0.05 decrease with value ranged from 12.34±21.3 to 75.13±4.2 when compared with the standard drug with the value of 99.0 ± 0.26 and the nitric oxide inhibition activity carried out showed a significant P0.05decrease with the value ranged froml9.80±7.06 to 60.66±3.09. The scavenging antioxidant activity was more in DPPI-1 than others. According to the test result, AST showed a significant P 0.05 decrease ranging from 32.00±1.13 to 69.45± 0.77 when compared with the negative group with value of 76.40± 0.56. ALT showed a significant P 0.05 decrease in value ranging from 3 1.20 ±0.56 to 68.30± 0.70 when compared with the negative group with value of 73.95± 0.77. Results obtained from TB showed a significant P 0.05 decrease with values ranging from 11.95±1.06 to 24.25 ± .0.91 when compared with the negative group value of 28.75± 0.77.Results of CB Showed a significantP0.05 decrease of values ranging from 4.10 ± 0.14 to 7.90±0.14 when compared with the negative group value of 9.55 ± 0.91.Urea result showed a significant P0.05decrease with values ranging from 40.40± 1.27 to 57.50± 0.98 when compared with the negative group value of 59.55± 1.20.Creatinine also showed a significant P0.05 decrease in groups 3-7 with values ranging from 3.55±0.07 to 5.85±0.07 when compared with the negative group value of 6.60 ± 0.28.Group 1 shows highest increase in all the parameters thereby indicating symptoms of liver and kidney damage in the groups. From the results, reduction in the biochemical parameters seen in group 3 — 7 indicated that Peniacleihra inacrophylla seed have hepatorenal protective activities and should be used in folk medicine to ameliorate acetaminophen (paracetamol) induced toxicity.
