                                                 ABSTRACT

Paracetamol could induce nephrotoxicity and even more severe fatal acute hepatic d mage when taken in an overdose. This study aims to evaluate the nephron protective potentials f vit C and vit E in paracetamol-induced nephrotoxicity The comparative nephron-protective potential of vit C and vit B against paracetamol-induced nephrotoxicity was assessed in this study. The vit C at piophylactic dosage were as follows, (25mg,50mg,100mg,150mg,200mg) and vitamin E (lOOiu,200iu,300iu,400iu,500iu) were separately administered orally to the rats (group 1-5). The protective potentials of ascorbic acid and a-tocopherol was significantly increased with increase in dosage co-administration of the synthetic antioxidants. Male albino Wister rat overdosed with paracetamol showed kidney damage and oxidative stress as indicated by significantly (P<O.05) increased, serum levels of creatinine total and urea. At the same time, there were decreased activities of serum creatinine total and urea level in all the treated groups with high dosage of vitamin C and vitamin B when compared with the control group. Treatment with Co-administration of Vitamin C and vitamin E doses of 50 and 40 mg/kg body weight could significantly (P<0.05) decrease the kidney damage marker enzymes, urea and crealinine levels when compared with paracetamol into indicated group. These results suggest that Co-administration of Vitamin C and vitamin E exhibits a potent nephron-protectively effect on paracetamol-induced kidney damage in rats, which may be due to both increased the antioxidant enzymes activity and inhibition of lipid peroxidation.
