ABSTRACT
The effect of methanol extract of Urtica dioica leaf on male hormone profile and prostate specific antigen (PSA) level in dihydrotestosterone- estradiol valarate induced with Benign Prostate Hyperplasia (BPH) was studied .Twenty four animals divided into five experimental groups namely normal control (NC), disease control, drug control (Finasteride), study group 1(loomg/kgbwt of Urtica dioica leaf extact), study group 2 (200mg/kg bwt of Urlica dioica leaf extract). Normal control group receive pure olive oil orally and subcutaneous injection every day. Disease control group and drug control group received subcutaneous injection of 0.9mg/kg of dihydrotestosterone (DHT) and olive oil and finasteride orally for 28 days respectively. Study group 1 and study group 2 received subcutaneous injection of 9mg/kg body weight of DHT and 100mg/kg and 200mg/kg of Urtica dioica leaf extract respectively for 28 days. Parameters analysed include prostate specific Antigen (PSA), male hormones (follicle stimulating hormone and luteinising hormone) and the prostate gland. The result showed that PSA levels decrease significantly (P<0.05) in groups treated with Urica dioica leaf extract (100mg/kg and 200mg/kg) compared to normal group and disease control group. Also the body weight of those treated with Urtica dioica leaf extract also increases. The result of the relative prostate weight showed significant (P<0.05) increase in disease control when compared with other experiment, whereas, there was no significant (P0<0.05) differences in the relative heart and kidney weight of the disease control when compared with other experiment. The result also showed a significant (p<O.05) increase in male hormones and testosterone of those treated with Urtica dioica leaf extract when compared with other experimental groups. These findings indicate that Urtica dioica leaf extract may prevent or suppress the enlargement of BPH and may be a useful phytotherapy in the treatment of BPH.
