ABSTRACT
Diabetes mellitus is a metabolic disorder of
glucose metabolism. The management of blood glucose level is the hallmark in
the treatment ofthis disease. This may be achieved through the use of oral
hypoglycemic drugs such as sulfonylureas, metformin, insulin secretagogues, and
alpha amylase, alpha glucosidase and pancreatic lipase inhibitors. The purpose
of the present study was to investigate • Anti-diabetic Screening potential of
methanol extract from Alchornea laxiflora and Dialium guineense against
carbohydrate hydrolyzing enzymes such as alpha amylase, alpha glucosidase and
pancreatic lipase. This was performed using porcine pancreatic alpha amylase
from Aspergillus oryzae, alpha glucosidase from Saccharomyces cerevisiae and
pancreatic lipase. Highest concentration of methanol extract ofA. laxiflora and
extract of D.guineense possesses the highest percentage inhibitory activity
against amylase (A.laxiflora Img/ml (60.87%± 0.724) , D.guineense Img/ml
(77.34% ±11.33), alpha glucosidase {A.laxiflora 0.2mg/ml (78.44% ± 0.29),
D.guineense 0.2mg/ml (66.55%±0.34)and pancreatic lipase (A.laxiflora Img/ml
(52.33%± 1.64), D.guineense Img/ml (54.73%±5.66)comparable to the standard
drugs, acarbose (80.00% ±0.73) and orlistat (60.03±1.35). It can be concluded
that methanol extracts of A.laxiflora . and D.guineense exhibited a potent
inhibitory activity on the three enzymes studied.
